The condition of your strings plays a part in the guitars tunablility, Strings that are old or have crud built up on them are the main culprits. If you hold your guitar sideways with a white wall as the background then you may be able to see 'crud' built up on the strings, Or if your strings start feeling 'catchy' rather than slick then its probably time to change them...
This 'gunk' is from your hands so its always best to clean your hands before you play and clean them off after you are done...

Even just a little bit of gunk on your strings will make the 'intonation'
out of whack...
If your strings aren't 'rusty' then you can probably use a cloth to clean each of the unwound strings, Wrap a small peice of cloth around the string and then slide it up the string. You should feel the gunk catching onto the cloth as you slide it up and down, You can probably even see the 'gunky' stuff falling off of the strings as you do it.
The 'wound' strings do not really have to be cleaned but still need to be changed when you change the other strings, The grooves in the wound strings will not be affected by gunk because it goes in the grooves but doesn't change the the string size (eww). The changing of string size is what makes it go out of tune or become unintonated, If you change the string size drastically then you will have to check the intonation. (Example: start out with '.047-.09's' and then change to '.052 - 11's')...
